Indications/Uses
Produces regression in acute leukaemias, non-Hodgkin's lymphoma, soft tissue & osteogenic sarcomas, & solid tumors eg, breast, lung, head & neck, bladder, cervical, ovarian & testicular carcinomas. Patient w/ severe psoriasis who are unresponsive to other forms of treatment.
Dosage/Direction for Use
Not to exceed 30 mg/m2 as single dose for not >5 successive days w/ 2-wk rest period between treatments.
Administration
Should be taken on an empty stomach: May be taken w/ meals to reduce GI discomfort. Avoid taking w/ milk-rich products.
Contraindications
Serious anaemia, leucopenia or thrombocytopenia. Severe renal or hepatic impairment.
Special Precautions
Alcoholics or w/ ulcerative GIT disorders. Monitor plasma conc & urinary excretion during high-dose regimens; CBC w/ platelet counts & liver function. Maintain adequate urine flow. Hepatic or renal impairment. Concurrent use of aminobenzoic acid, chloramphenicol, phenylbutazone, phenytoin, probenecid, salicylates, sulphonamides & tetracyclines; NSAIDs. May impair fertility. Not to be administered during pregnancy & lactation.
Adverse Reactions
Leucopenia, thrombocytopenia, anaemia, mouth ulceration, stomatitis, diarrhea. Bone marrow depression, megaloblastic anaemia. Hypogammaglobulinaemia. Liver & kidney damage. Osteoporosis, effects on skin & nails, pulmonary reactions including interstitial pneumonitis. Metabolic alterations; neurotoxic reactions; teratogenic effects.
Drug Interactions
Enhanced effects w/ aminobenzoic acid, chloramphenicol, phenylbutazone, phenytoin, probenecid, salicylates, sulfonamides & tetracyclines. Fatal toxicity w/ NSAIDs.
